The mechanism of action of Praventin for the internal support of healthy skin is not clear; it is known that part of the bioactive protein complex contains lactoferrin. Lactoferrin is present naturally in exocrine secretions that are commonly exposed to normal flora: milk, tears, nasal exudate, bronchial mucus, gastrointestinal fluids, and saliva. Lactoferrin is expressed in polymorphonuclear leukocytes and is secreted by various circulating immune cells at infected sites. A primary function of lactoferrin is scavenging free iron in body fluids and inflamed sites to suppress free radical-mediated damage and diminish availability of the element to invading microbes. Additionally, bovine lactoferrin possesses anti-microbial, anti-inflammatory and anti-oxidant properties which are relevant to the support of a healthy complexion.

Clinical Studies on Praventin

Praventin inhibits the growth of propionibacterium acnesPraventin may limit the growth of acne bacteria

Propionibacterium acnes (ATCC 11827) was cultivated in reinforced clostridial medium at 37ºC in an anaerobic chamber under an atmosphere of 90% N2, 5% CO2 and 5% H2. Bacteria were suspended at a concentration range of 10e6 CFU/ ml in reinforced clostridial medium. 16% Praventin solutions were made in sterile water. In 96 wells microplates, 100ul bacterial suspension was added to 50µl Praventin solution and 50ul 2* concentrated reinforced clostridial medium. OD (600 nm) was measured at 24, 36, 48, 60 and 72 hours.

In figure 4 it is shown that Praventin inhibits the growth of Propionibacterium acnes. It is possible that lactoferrin may contribute to the anti-microbial properties of Praventin against propionibacteria or other acne-related bacteria. This may be accomplished by mechanisms such as ironscavenging, disruption of cell membrane or inhibition of biofilm formation.

The lactoferrin fraction in Praventin may exert anti-inflammatory effects in the skin by skewing immune response in the anti-inflammatory direction. This is initiated through binding of free iron, which catalyzes formation of free radicals. Praventin may neutralize the release of the inflammation mediator lipopolysaccharides (LPS) by acne-related bacteria, or modulate the activity of immune cells. The reduced redness in the consumer studies is indicative for an anti-inflammatory effect of Praventin.

 

Praventin assists in the inhibition of squalene oxidation

In a study with 10 healthy volunteers, it was demonstrated that topical application of Praventin was able to prevent cutaneous lipid peroxidation

 

Praventin may exert activity by suppressing the oxidation of squalene (a compound of skin lipids). In previous studies, the lipids found on the skin surface, and from open and closed comedones in acne patients were analysed. It was demonstrated that these lipids were enriched in polar lipids, as compared with skin surface lipids acquired from acne-free controls. In both open and closed comedones, these polar lipids appeared to be derived mainly from the oxidation of squalene. It was suggested that squalene oxidation is the link between comedogenesis and bacterial colonization, and plays an essential role in the pathogenesis of acne. In a study with 10 healthy volunteers, it was demonstrated that topical application of Praventin was able to prevent cutaneous lipid peroxidation, as measured by the squalene: squalenehydroperoxide ratio (unpublished results). These results indicate that Praventin may support a healthy complexion by preventing squalene oxidation, and subsequent comedogenesis.
